Objective

To determine the solubility of UCExp251-2C from UCExp251 (UC148B) in a) Benzene, b) THF, c) acetonitrile, d) methanol and e) ethanol

Procedure

To five labeled half dram vials, a small amount of UCExp251-2C was added to each vials containg a) Benzene, b) THF, c) acetonitrile, d) methanol and e) ethanol. Put them into Lauda heating/cooling device at 40C in half an hour, and cool them dowm at 25C in half an hour. Proton HNMR spectra of supernatant from each solution was obtained on a 500MHz Varian invo instrument. Solubility was determined by using SAMS.

17:40 Turned on a "Lauda heating/cooling" device. Set up temperature to exact 40C
17:45 Took out 5 half-dram vials, labeled ONSC145-1A, ONSC145-2A, ONSC145-3A, ONSC145-4A and ONSC145-5A. Put 200ul of benzene into vial 1, THF into vial 2, acetonitrile into vial 3, methanol into vial 4, ethanol into vial 5.
17:50 Took out small mount of solid UCExp251-2C into each vial. Vortexed each vials carefully to make sure there is still solid remained in each vials. Parafilmed carefully all the vials. Took a photo of 5 vials before putting into the bath.
17:55 Put 5 vials into "Lauda heating/cooling" device. At this time, the temperature in the bath was 40C.
18: 25 Lowered the bath temperature to 25C.
18:55 The temperature in the bath reached 25C

11:20 Removed the samples from the 25C bath and centrifuged the vials.
11:30 Pipetted out 700uL CDCl3 in to five labled one dram vials
11:40 Carefully pipetted out 100uL of the supernatants

Supernatant from the half dram vial ONSCExp145-5A in to the one dam vial; ONSCExp145-5B, vortexed the resulting solution and placed them in to five clean and dry NMR tubes.
