Objective

To study the effect of phenylacetic acid on the solubility of the Ugi product UCExp17C at lower concentration (0.01M - 0.15) in benzene

Procedure

154A- 0.5M: Dissolve phenylacetic acid (1mmol, 136mg) in benzene in a 2mL volumetric flask - Stock solution
154B - 0.2M - Pipette out 400uL of 154A in to a 1mL volumetric flask and make it up to the mark with benzene
154C - 0.1M - Pipette out 200uL of 154A in to a 1mL volumetric flask and make it up to the mark with benzene
154D - 0.075M - Pipette out 150uL of 154A in to a 1mL volumetric flask and make it up to the mark with benzene
154E - 0.05M - Pipette out 100uL of 154A in to a 1mL volumetric flask and make it up to the mark with benzene
154F - 0.03M - Pipette out 60uL of 154A in to a 1mL volumetric flask and make it up to the mark with benzene
154G - 0.01M- Pipette out 20uL of 154A in to a 1mL volumetric flask and make it up to the mark with benzene

18:15 Weighed out 136mg of phenylacetic acid and put into a 2ml volumetric flask. Filled up to the mark with benzene. This is the stock solution-154A-0.5M
18:20 Transferred stock solution into a dram vial to store it in the refrigerator to prevent evaporation.
[this won't be an issue for benzene boiling at 80C JCB]- [ok - Hai]

11:10 Changed the temperature setting of the water-glycol to 25C.

17:15 Put the stock solution in 25C water for 20 minutes.
17:35 Took out six 1ml-volumetric flask from the hot oven. Labeled B,C,D,E,F, and G.
In volumetric flask B: pipetted out 400uL of 154A into flask B and make it up to the mark. This solution is 0.2M
In volumetric flask C: pipetted out 200uL of 154A into flask C and make it up to the mark. This solution is 0.1M
In volumetric flask D: pipetted out 150uL of 154A into flask D and make it up to the mark. This solution is 0.075M
In volumetric flask E: pipetted out 100uL of 154A into flask E and make it up to the mark. This solution is 0.05M
In volumetric flask F: pipetted out 60uL of 154A into flask F and make it up to the mark. This solution is 0.03M
In volumetric flask G: pipetted out 20uL of 154A into flask G and make it up to the mark. This solution is 0.01M
17:45 Took out 7 half dram vials. Labeled Exp154-A, 154-B, 154-C, 154-D, 154-E, 154-F, and 154-G.
In vial Exp154-A: pipetted out 500uL out of stock solution 154-A into this vial. Put small amount of Ugi176C so that there was still small amount of solid remain in the solution.
In vial Exp154-B: pipetted out 500uL out of flask B into this vial. Put small amount of Ugi176C so that there was still small amount of solid remain in the solution.
In vial Exp154-C: pipetted out 500uL out of flask C into this vial. Put small amount of Ugi176C so that there was still small amount of solid remain in the solution.
In vial Exp154-D: pipetted out 500uL out of flask D into this vial. Put small amount of Ugi176C so that there was still small amount of solid remain in the solution.
In vial Exp154-E: pipetted out 500uL out of flask E into this vial. Put small amount of Ugi176C so that there was still small amount of solid remain in the solution.
In vial Exp154-F: pipetted out 500uL out of flask F into this vial. Put small amount of Ugi176C so that there was still small amount of solid remain in the solution.
In vial Exp154-G: pipetted out 500uL out of flask G into this vial. Put small amount of Ugi176C so that there was still small amount of solid remain in the solution.
18:10 Parafilmed all the vials
18:30 Put all vial into the Lauda heating/cooling bath. The temperature in the bath was 50C.
19:35 Adjusted temperature in the bath to 40C.

17:55 Changed the temperature setting on the bath from 40C to 25C. Checked and made sure all the sample vials appeared ok-no leak!

11:20 Removed the vials from the temperature control bath. All vials contained solvent along with solid in them.
11:30 Centrifuged the vial.
11:35 Pipetted out 400uL DMSO-d6 in to four clean and dry MR tubes. Labeled them ONSCExp154-A-1 through ONSCExo154-G-1.
11:55 Carefully pipetted out approximately 200uL supernatant solution from each vial and transferred it in to a corresponding NMR tube.
14:45 Obtained HNMR of the solution- HNMRs labeled ONSCExp154-A-1 to ONSCExp154-G-1.
